RESUMEN

The traceability of in vitro diagnostics or drug products is based on the accurate quantification of proteins. In this study, we developed an absolute quantification approach for proteins. This method is based on calibrated particle counting using electrospray-differential mobility analysis (ES-DMA) coupled with a condensation particle counter (CPC). The absolute concentration of proteins was quantified with the observed protein particle number measured with ES-DMA-CPC, and the detection efficiency was determined by calibrators. The measurement performance and quantitative level were verified using two certificated reference materials, BSA and NIMCmAb. The linear regression fit for the detection efficiency values of three reference materials and one highly purified protein (myoglobin, BSA, NIMCmAb and fibrinogen) indicated that the detection efficiency and the particle size distribution of these proteins exhibited a linear relationship. Moreover, to explore the suitability of the detection efficiency-particle size curve for protein quantification, the concentrations of three typical proteinaceous particles, including two high molecular weight proteins (NIST reference material 8671 and D-dimer) and one protein complex (glutathione S-transferase dimer), were determined. This work suggests that this calibrated particle counting method is an efficient approach for nondestructive, rapid and accurate quantification of proteins, especially for measuring proteinaceous particles with tremendous size and without reference standards.

A correlation exists between obstructive sleep apnoea (OSA) and the severity of metabolic dysfunction-associated steatotic liver disease (MASLD), OSA can induce more severe MASLD. However, the underlying regulatory mechanism between the two is unclear. To this end, this study explored the role and possible molecular mechanisms of adipocyte-derived exosomes under OSA in aggravating MASLD. Through sequencing technology, miR-455-3p was identified as a co-differentially expressed miRNA between the MASLD + OSA and Control groups and between the MASLD + OSA and MASLD groups. Upregulation of TCONS-00039830 and Smad2 and downregulation of miR-455-3p in the MASLD and MASLD + OSA groups were validated in vivo and in vitro. TCONS-00039830, as a differentially expressed LncRNA in exosomes found in the sequencing results, transfection notably downregulated miR-455-3p and upregulated Smad2 in hepatocytes. TCONS_00039830 overexpression increased fat, triglyceride and cholesterol levels, while miR-455-3p overexpression decreased these levels. Furthermore, exosome administration promoted the accumulation of fat, triglyceride and cholesterol, upregulated TCONS_00039830 and Smad2, and downregulated miR-455-3p. Overexpression of miR-455-3p reversed the increased fat accumulation and upregulated TCONS_00039830 and Smad2. In conclusion, OSA-derived exosomes promoted hepatocyte steatosis by regulating TCONS_00039830/miR-455-3p/Smad2 axis, thereby aggravating liver damage in MASLD.

Objective: To explore risk factors of electrical status epilepticus during sleep in children with benign childhood epilepsy with centro-temporal spikes (BECT). Methods: This is a clinical comparative study. The subjects of study were 67 children with BECT from the Outpatient Department of Pediatric Neurology in Xingtai People's Hospital from January 2019 to January 2022. According to the occurrence of ESES, the enrolled children were divided into control group which included BECT children without ESES and the observation group which included BECT children with ESES. Compared differences of the two groups in the age of first seizure, the frequency of seizures before treatment, the classification of treatment drugs, cranial MRI, and discharge side of electroencephalogram (EEG). Results: There was no statistical difference between the two groups in the frequency of seizures before treatment, the classification of treatment drugs, cranial MRI, and the distribution of EEG discharges in the left and right cerebral areas(P>0.05). Statistical differences were observed in the age of the first seizure, whether the seizures occurred after treatment, and EEG discharges in unilateral/bilateral cerebral areas (P<0.05). Furthermore, the collinearity test and Logistic regression analysis showed that the age of the first seizure, the frequency of seizures before treatment, and whether the seizures occurred after treatment were independent risk factors for the occurrence of ESES in BECT (P<0.05). Conclusion: Clinically, the occurrence of ESES in children with BECT may be related to the younger age of the first seizure, higher frequency of seizures before treatment, and the occurrence of seizures after treatment.

This paper reports a facile fabrication method of hydroxyapatite/chitosan (HAp/CS) composite scaffold with 3D porous structure without using any chemical cross-linkers. The HAp particles had an urchin-like hollow microstructure and high surface area, which was uniformly dispersed into the pore walls of the HAp/CS scaffold. The addition of HAp can efficiently enhance the mechanical properties and bioactivity of the HAp/CS scaffold. Moreover, periostin was successfully loaded onto the HAp/CS scaffold. When applied to the repair of bone defect in a rat mandibular model, the HAp/CS scaffold loaded with periostin can enhance osteointegration and accelerate bone regeneration. Our research combines periostin with the HAp/CS composite material, which provides a novel strategy to improve bone regeneration and has great application prospect in bone repair fields.

The solar-driven photocatalytic production of hydrogen peroxide (H2 O2 ) from water and oxygen using semiconductor catalysts offers a promising approach for converting solar energy into storable chemical energy. However, the efficiency of photocatalytic H2 O2 production is often restricted by the low photo-generated charge separation, slow surface reactions and inadequate stability. Here, we developed a mixed-linker strategy to build a donor-acceptor-acceptor (D-A-A) type covalent organic framework (COF) photocatalyst, FS-OHOMe-COF. The FS-OHOMe-COF structure features extended π-π conjugation that improves charge mobility, while the introduction of sulfone units not only as active sites facilitates surface reactions with water but also bolsters stability through increased interlayer forces. The resulting FS-OHOMe-COF has a low exciton binding energy, long excited-state lifetime and high photo-stability that leads to high performance for photocatalytic H2 O2 production (up to 1.0 mM h-1 ) with an H2 O2 output of 19 mM after 72 hours of irradiation. Furthermore, the catalyst demonstrates high stability, which sustained activity over 192 hours of photocatalytic experiment.

The partial pressure of ocean surface CO2 (pCO2) plays an important role in quantifying the carbon budget and assessing ocean acidification. For such a vast and complex ocean system as the global ocean, most current research practices tend to study the ocean into regions. In order to reveal the overall characteristics of the global ocean and avoid mutual influence between zones, a holistic research method was used to detect the correlation of twelve predictive factors, including chlorophyll concentration (Chlor_a), diffuse attenuation coefficient at 490 nm (Kd_490), density ocean mixed layer thickness (Mlotst), eastward velocity (East), northward velocity (North), salinity (Sal), temperature (Temp), dissolved iron (Fe), dissolved silicate (Si), nitrate (NO3), potential of hydrogen (pH), phosphate (PO4), at the global ocean scale. Based on measured data from the Global Surface pCO2 (LDEO) database, combined with National Aeronautics and Space Administration (NASA) Ocean Color satellite data and Copernicus Ocean reanalysis data, an improved optimized random forest (ORF) method is proposed for the overall reconstruction of global ocean surface pCO2, and compared with various machine learning methods. The results indicate that the ORF method is the most accurate in overall modeling at the global ocean scale (mean absolute error of 6.27µatm, root mean square error of 15.34µatm, R2 of 0.92). Based on independent observations from the LDEO dataset and time series observation stations, the ORF model was further validated, and the global ocean surface pCO2 distribution map of 0.25° × 0.25° for 2010 to 2019 was reconstructed, which is of significance for the global ocean carbon cycle and carbon assessment.

As an advanced sensing technology, dual-mode flexible sensing, integrating both tactile and touchless perception, propels numerous intelligent devices toward a more practical and efficient direction. The ability to incorporate multiple sensing modes and accurately distinguish them in real time has become crucial for technological advancements. Here, we proposed a dual-mode sensing system (B-MIGS) consisting of a dual-layer sensing device with a magnetically induced grid structure and a testing device. The system was capable of utilizing mechanical pressure to perceive tactile stimulation and magnetic sensing to simultaneously transduce touchless stimulation simultaneously. By leveraging the triboelectric effect, the decoupling of tactile and touchless signals in the presence of unknown signal sources was achieved. Additionally, the sensing characteristics of the B-MIGS were optimized by varying the curing magnetic induction intensity and magnetic particle concentration. The influence of the temperature and humidity on the sensing signals was also discussed. Finally, the practical value of the B-MIGS as a dual-mode monitoring system was demonstrated on soft petals and sensor arrays, along with exploration of its potential application in underwater environments.

Nanohydrogelation of covalent organic frameworks (COFs) will undoubtedly open up new applications for them in water, such as aqueous catalysis and biomedicine. It is currently a great challenge to achieve water dispersion of COFs through either bottom-up construction strategies or top-down exfoliating technologies. Herein, poly(N-isopropylacrylamide) (PNIPAM)-postmodified COF nanohydrogels (COF-NHGs) are successfully designed and synthesized via in situ atom-transfer radical polymerization (ATRP) on a scaffold of COFs. During the polymer growth process, the bulk COFs are exfoliated into nanosheets with a lateral size of ∼500 nm and a thickness of ∼6.5 nm. Moreover, their size can be precisely controlled by the degree of polymerization of PNIPAMs. In aqueous solution, the obtained COF-NHGs are assembled into nanohydrogels retaining intra-plane crystallinity and exhibit a temperature-sensitive sol-gel phase transition. With excellent solubility in organic solvents, the COF-NHGs' intrinsic physical properties in the solution state can be characterized through their solution nuclear magnetic resonance and ultraviolet absorption spectra. These results put forward new opportunities for regulating the solution processability of COFs and building an intelligent, stimuli-response platform of COF-polymer composite nanohydrogels for device applications.

The manipulation of electronic structure and prevention of photogenerated carriers from being quenched in bulk defects during the photocatalytic CO2 reduction reaction (CRR) have been effectively demonstrated through surface vacancy and defect engineering. In this work, the electronic structure on the surface of Zn3In2S6/ZnS (ZIS/ZnS) is significantly modified by the introduction and control of the surface S vacancies (SV) through Ar-plasma treatment. EPR and XPS analyses confirmed that SV was exclusively present on the ZIS/ZnS surface. The resulting ZIS/ZnS heterojunction photocatalysts demonstrate an impressive 46.6% selectivity toward C2 products even in the absence of cocatalysts. The mechanism of photocatalytic CRR is further elucidated through in situ analysis. Theoretical calculations demonstrate that the presence of In and Zn atoms adjacent to SV significantly enhances the adsorption of CO2 and facilitates C-C coupling.

BACKGROUND: Severe community-acquired pneumonia (SCAP) results in high mortality as well as massive economic burden worldwide, yet limited knowledge of the bio-signatures related to prognosis has hindered the improvement of clinical outcomes. Pathogen, microbes and host are three vital elements in inflammations and infections. This study aims to discover the specific and sensitive biomarkers to predict outcomes of SCAP patients. METHODS: In this study, we applied a combined metagenomic and transcriptomic screening approach to clinical specimens gathered from 275 SCAP patients of a multicentre, prospective study. FINDINGS: We found that 30-day mortality might be independent of pathogen category or microbial diversity, while significant difference in host gene expression pattern presented between 30-day mortality group and the survival group. Twelve outcome-related clinical characteristics were identified in our study. The underlying host response was evaluated and enrichment of genes related to cell activation, immune modulation, inflammatory and metabolism were identified. Notably, omics data, clinical features and parameters were integrated to develop a model with six signatures for predicting 30-day mortality, showing an AUC of 0.953 (95% CI: 0.92-0.98). INTERPRETATION: In summary, our study linked clinical characteristics and underlying multi-omics bio-signatures to the differential outcomes of patients with SCAP. The establishment of a comprehensive predictive model will be helpful for future improvement of treatment strategies and prognosis with SCAP. The progress of solar-driven water-splitting technology has been impeded by the limited light response capability of semiconductor materials. Despite attempts to leverage nearly 50% of infrared radiation for photothermal synergy and catalytic reaction enhancement, heat loss during liquid phase reactions results in low energy conversion efficiency. Here, the photothermally driven catalytic water-splitting system, which designs K-SrTiO3 -loaded TiN silica wool at the water-air interface. Photocatalytic tests and density functional theory calculations demonstrate that the thermal effect transforms liquid water into water vapor, thereby reducing the reaction free energy of catalysts and improving the transmission rate of catalytic products. Hence, the hydrogen evolution rate reaches 275.46 mmol m-2  h-1 , and the solar-to-hydrogen (STH) efficiency is 1.81% under 1 sun irradiation in this gas-solid system, which is more than twice that of liquid water splitting. This novel photothermal catalytic pathway, which involves a coupled reaction of water evaporation and water splitting, is anticipated to broaden the utilization range of the solar spectrum and significantly enhance the conversion efficiency of STH.

A three-dimensional rigid "six-connected" porous triptycene network based on Tröger's base (TB-PTN) was synthesized by using triptycenes as connectors and Tröger's base as linkers. With characteristics of a high surface area of 1528 m2 g-1, nitrogen-enriched groups, and superior thermal stability, TB-PTN displays a high CO2 uptake of 22.3 wt % (273 K, 1 bar) and excellent iodine vapor adsorption (240 wt %).

Photocatalytic hydrogen evolution through water splitting offers a promising way to convert solar energy into chemical energy. Covalent triazine frameworks (CTFs) are ideal photocatalysts owing to its exceptional in-plane π-conjugation, high chemical stability, and sturdy framework structure. However, CTF-based photocatalysts are typically in powder form, which presents challenges in catalyst recycling and scale-up applications. To overcome this limitation, we present a strategy for producing CTF films with excellent hydrogen evolution rate that are more suitable for large-scale water splitting due to their ease of separation and recyclability. We developed a simple and robust technique for producing CTF films on glass substrates via in-situ growth polycondensation, with thicknesses adjustable from 800 nm to 27 µm. These CTF films exhibit exceptional photocatalytic activity, with the hydrogen evolution reaction (HER) performance reaching as high as 77.8 mmol h-1 g-1 and 213.3 mmol m-2 h-1 with co-catalyst Pt under visible light (≥420 nm). Additionally, they demonstrate good stability and recyclability, further highlighting their potential in green energy conversion and photocatalytic devices. Overall, our work presents a promising approach for producing CTF films suitable for a range of applications and paves the way for further developments in this field.

Recently, photo switching porous materials have been widely reported for low energy costed CO2 capture and release via simply remoted light controlling method. However, most reported photo responsive CO2 adsorbents relied on metal organic framework (MOFs) functionalisation with photochromic moieties, and MOF adsorbents still suffered from chemically and thermally unstable issues. Thus, further metal free and highly stable organic photoresponsive adsorbents are necessary to be developed. CTFs, because of their high porosity and stability, have attracted great attention for CO2 capture. Considering the high CO2 uptake capacity and structural tunability of CTFs, it suggests high potential to fabricate the photoswitching CTF materials by the same functionalisation method as MOFs. Herein, the first series of photo switching CTFs were developed for low energy CO2 capture and release. Apart from that, the CO2 switching efficiency could be doubled either through the azobenzene numbers adjusting method or through the previously reported structural alleviation strategy. Furthermore, the pore size distribution of azobenzene functionalised PCTFs also could be tuned under UV exposure, which may contribute to the UV light induced decrease of CO2 uptake capacity. These photoswitching CTFs represented a new kind of porous polymers for low energy costed CO2 capture.

Not from concentrate (NFC) orange juice is minimally processed, natural-appearing food, and has become more popular. Sterilization is an important stage for NFC orange juice production. Here we present a comprehensive analysis of the effect of sterilization on the metabolites of NFC orange juices, including three thermal (pasteurization, high-temperature short time, and ultra-high temperature) and one nonthermal (high hydrostatic pressure) method. A total of 108 metabolites, including 59 volatiles and 49 nonvolatiles were identified in orange juice. Of which, only butyl butanoate and 3-carene were detected in fresh orange juice. Sterilization significantly changed the metabolites of orange juice, and different methods caused various changes. Esters were downregulated by both thermal and nonthermal sterilization, while most flavonoids and terpenes were upregulated. With comparative analyses of three thermal sterilization, we found that high temperature and relatively short-time treatment preserved esters and ascorbic acid more effectively than low temperature and prolonged treatment. Aldehydes, however, were the opposite. Nonthermal sterilization is effective in preserving the metabolites of orange juice, especially for esters, terpenes, and flavonoids. In addition, 19 distinct metabolites were characterized between thermal and nonthermal samples by chemometrics analysis. These findings provide a new sight of the optimization of sterilization methods and references for different types of NFC orange juice identification. PRACTICAL APPLICATION: This study provides a reference for the optimization of sterilization methods and identification of HHP and thermal NFC orange juice and also benefits the purchase of consumers.

Metagenomic sequencing (mNGS) is a powerful diagnostic tool to detect causative pathogens in clinical microbiological testing owing to its unbiasedness and substantially reduced costs. Rapid and accurate classification of metagenomic sequences is a critical procedure for pathogen identification in dry-lab step of mNGS test. However, clinical practices of the testing technology are hampered by the challenge of classifying sequences within a clinically relevant timeframe. Here, we present GPMeta, a novel GPU-accelerated approach to ultrarapid pathogen identification from complex mNGS data, allowing users to bypass this limitation. Using mock microbial community datasets and public real metagenomic sequencing datasets from clinical samples, we show that GPMeta has not only higher accuracy but also significantly higher speed than existing state-of-the-art tools such as Bowtie2, Bwa, Kraken2 and Centrifuge. Furthermore, GPMeta offers GPMetaC clustering algorithm, a statistical model for clustering and rescoring ambiguous alignments to improve the discrimination of highly homologous sequences from microbial genomes with average nucleotide identity >95%. GPMetaC exhibits higher precision and recall rate than others. GPMeta underlines its key role in the development of the mNGS test in infectious diseases that require rapid turnaround times. Further study will discern how to best and easily integrate GPMeta into routine clinical practices. GPMeta is freely accessible to non-commercial users at https://github.com/Bgi-LUSH/GPMeta.

Metabolites including antibiotics, enzymes, and volatiles produced by plant-associated bacteria are key factors in plant-microbiota interaction that regulates various plant biological processes. There should be crucial mediators responsible for their entry into host plants. However, less is known about the identities of these plant transporters. We report that the Arabidopsis Nitrate Transporter1 (NRT1)/NPF protein NPF2.13 functions in plant uptake of tunicamycin (TM), a natural antibiotic produced by several Streptomyces spp., which inhibits protein N-glycosylation. Loss of NPF2.13 function resulted in enhanced TM tolerance, whereas NPF2.13 overexpression led to TM hypersensitivity. Transport assays confirmed that NPF2.13 is a H+ /TM symporter and the transport is not affected by other substrates like nitrate. NPF2.13 exclusively showed TM transport activity among tested NPFs. Tunicamycin uptake from TM-producing Streptomyces upregulated the expression of nitrate-related genes including NPF2.13. Moreover, nitrate allocation to younger leaves was promoted by TM in host plants. Tunicamycin could also benefit plant defense against the pathogen. Notably, the TM effects were significantly repressed in npf2.13 mutant. Overall, this study identifies NPF2.13 protein as an important TM transporter in plant-microbe interaction and provides insights into multiple facets of NPF proteins in modulating plant nutrition and defense by transporting exterior bacterial metabolites.

Covalent Triazine Frameworks (CTFs) have received great attention from academia owing to their unique structure characteristics such as nitrogen-rich structure, chemical stability, fully conjugated skeleton and high surface area; all these unique properties make CTFs attractive for widespread applications, especially for photocatalytic applications. In this review, we aim to provide recent advances in the CTFs preparation, and mainly focus on their photocatalytic applications. This review provides a comprehensive and systematic overview of the CTFs' synthetic methods, crystallinity lifting strategies, and their applications for photocatalytic water splitting. Firstly, a brief background including the photocatalytic water splitting and crystallinity are provided. Then, synthetic methods related to CTFs and the strategies for enhancing the crystallinity are summarized and compared. After that, the general photocatalytic mechanism and the strategies to improve the photocatalytic performance of CTFs are discussed. Finally, the perspectives and challenges of fabricating high crystalline CTFs and designing CTFs with excellent photocatalytic performance are discussed, inspiring the development of CTF materials in photocatalytic applications.

RESUMEN

Due to its excellent bone conductivity and drug adsorption as well as pH-responsive drug release property, hydroxyapatite (HAp) is widely used as a drug carrier in bone repair field. Here, we report for the first time a novel multi-functional polydopamine (PDA) coated Cu/F-codoped HAp (Cu/F-HAp-PDA) hollow microspheres. Both Cu2+ and F- were successfully doped into the lattice of HAp and uniformly distributed in the shell of hollow microspheres through a one-step hydrothermal synthesis. Then PDA was coated homogeneously on the outer layer of Cu/F-HAp hollow microspheres. Both Cu/F-HAp and Cu/F-HAp-PDA samples displayed high drug loading efficiency and pH responsive drug release behavior. Moreover, the obtained Cu/F-HAp-PDA hollow microspheres exhibited excellent photothermal conversion efficiency and photothermal stability. The molecular dynamics simulations showed that PDA and HAp can form mutual binding mainly through Ca-O bonding, while doxorubicin (DOX) is mainly bound to PDA molecules through hydrogen bonding and π-π stacking interaction.
